LTA COMMEFCIALTESTING & ENGINEEHINGGO.
METHODS co>mt.
The following tests were conducted in the Preliminary Toxicity Identification evaluation:
Dav 1
Initial Tonicity Test - D. magna were exposed to 0%, 1.9%, 25%, 50%, 75%, and 100%
effluent concentrations prepared using reconstituted laboratory water at 220 mg/L CaCOa.
Twenty organisms were exposed at each increasing concentration level.
Dav 2
Baseline Degradation Test - D. magna were exposed to a dilution water blank and 50%, 75%,
and 100% effluent concentrations prepared using reconstituted laboratory water at 220 mg/L
CaCOs. Twenty organisms per concentration were exposed approximately 24 hours after the start
of the Initial Toxicity Test.
Filtration/pH Adjustment Test (pH 3) -Twenty D. magna were exposed to a dilution water
blank and 50%, 75%, and l00% effluent adjusted to pH 3 with Hydrochloric Acid (HCl) and
filtered through a 0.45 µm fiber filter. Prior to organism exposure the effluent was readjusted
back to pHi° with Sodium Hydroxide (NaOH). Dilutions were prepazed with reconstituted
laboratory water at 220 mg/L CaCO,.
Filtration/pH Adjustment Test (pH 10) -Twenty D. magna were exposed to a dilution water
blank and 50%, 75%, and 100% effluent adjusted to pH 10 with Sodium Hydroxide (NaOH) and
filtered through a 0.45 µm fiber filter. Prior to organism exposure the effluent was readjusted
back to pHi with Hydrochloric Acid (IICI). Dilutions were prepared with reconstituted
laboratory water at 220 mg/L CaC03
Filtration/pH Unadjustment Test (pHi) -Twenty D. magna were exposed to a dilution water
blank and 50%, 75%, and 100% effluent without pH adjustment and filtered through a 1.0
micron glass fiber filter. Dilutions were prepared with reconstituted laboratory water at 220 mg/L
CaC03.
Aeration/pH Adjustment Test (pH 3) -Twenty D. magna were exposed to a dilution water
blank and 50%, 75%, and 100% effluent adjusted to pH 3 with Hydrochloric Acid (HCl) and
vigorously aerated for one hour. Prior to organism exposure the effluent was readjusted back to
pHi with Sodium Hydroxide (NaOH). Dilutions were prepared wish reconstituted laboratory
water at 220 mg/L CaC03.
° Initial pH
3
-526
