Ceriodaphnia dubia acute bioassay: <br />Dav 0 <br />1. At the end of the work day, the pH of the adjusted test <br />blank sample and the pH adjusted effluent sample is checked. <br />Acid and base are used to make final adjustments if <br />necessary. <br />2. Twenty organisms are exposed to five concentrations of pH <br />adjusted effluent, 1000, 500, 250, 12.5% and lo, prepared <br />using reconstituted laboratory water. Control replicates are <br />exposed to 100% reconstituted laboratory water and 100$ whole <br />effluent. Blank replicates are exposed to pH adjusted <br />laboratory water, 1000 sample. <br />Dav 1 - Solution Renewal <br />3. The pH of the manipulated laboratory water and the pH <br />adjusted 100% effluent samples are checked and re-adjusted, if <br />necessary, to pH i. Volumes used fer adjustments are <br />recorded. <br />4. Dilutions are prepared as above and the organisms are <br />transferred into the renewal solutions. <br />5.
