Task: Replicate technique using microsatellite primers used by Dr. Tom <br />Dowling in RBS evaluation, and develop a suite of additional microsatellite <br />primers. <br />Target date: July 2004 <br />C. Task: Replicate technique used in mtDNA RFLP analysis used by Dr. Tom <br />Dowling in screening RBS. <br />Target date: October 2004 <br />D. Task: Produce baseline genotype of all RBS broodstock at Dexter and other <br />facilities. <br />Target date: July 2005 <br />E. Task: Provide statistical analysis of results to determine the extent of <br />differentiation of hatchery stocks. <br />Target date: December 2005 <br />Alternative: If broodstocks appear to be distinct, such as Lake Mohave and the <br />Green River stocks, separate stock identities will be designated in collaboration <br />with the holding facilities, and mating strategies developed to ensure genetic <br />integrity is maintained. <br />Alternative: If stocks are not significantly different, movement of gametes and fry <br />between hatcheries will be part of ongoing broodstock management strategy to <br />maximize diversity of captive populations. <br />2) Establish a genetic broodstock plan that actively monitors and maintains genetic variability and <br />long-term fidelity of each broodstock at Dexter NFHTC. <br />Management Action: prevent loss of genetic diversity as measured by microsatellite loci, <br />and mtDNA. <br />32
