River included: at or near Goulds Wash, Ash Creek, and La Verkin Creek. It is unlikely
that these samples contained any fish stocked from Dexter because there are two water
diversion dams that serve as barriers and obstruct movement of chub between the
stocking site and sample site.
Extraction of DNA and Genotyping
Genomic DNA was extracted from fin clips using Qiagen DNeasy'e 96 Blood &
Tissue Kits following the manufacturer's instructions, and stored at -77°C. Nine
tetranucleotide microsatellite markers were used. in this study: Gel-227, CypG2, CypG5,
CypG9, CypG10, CvpG22, CvpG28, CypG30, and CypG48. Polymerase chain reaction
(PCR) primers for Gel 227 were developed by Keeler-Foster et al. (2004), and primers
for the remaining loci were developed by Baerwald and May (2004). Each 10 gl PCR
reaction contained the following final concentrations: 0.875u AmpliTaq Gold® DNA
polymerase (Applied Biosystems or AB); 1X GeneAmp® IOX PCR buffer; 2.5mM
