AVAILABLE PHOSPHOP.US IN MINE SOILS <br />(Modified method of Olsen, et a1, 1954) <br />Solutions: <br />1. Sodium bicarbonate (NaHCO ) solution, 0.519, adjusted to pH <br />8.5 with 1 M NaOH. Mineral oil added to avoid exposure to <br />the air; stored in a polyethylene container and made fresh <br />every 2 months. <br />2. Ammonium molt'§date, (NH ) Mo O 4 4H O, solution: <br />Dissolve l.5 g• {n 300 m~ ~f w~r~ disti~led and deionized <br />water. Filter if a.7.oudy and allow to cool. Gradually <br />add 342 ml of conc. HCL and mix. Dilute to 1 liter. <br />3. Concentrated SnCl 2H O solution: 10 g. of large crystals <br />dissolved in 25 m~. conc. HC1. Store in a brown glass <br />bottle in a refrigerator. Prepare fresh every Z months. <br />a) pilute SnCl add 0.5 ml of the conc. SnCl solution <br />to 66 ml. distilled and deionized water. 2Prepare <br />the dilute solution for each set of determinations. <br />• 4. Standard P solution: Weigh 0.4393 g. KH2PO4 into a 1 liter <br />volumetric flask. Add 500 mis. distilled and deionized <br />water and dissolve the salt. Dilute to 1 liter, and add <br />5 drops of toluene to reduce microbial growth. <br />5. Dilute P solution: pilute 20 ml. (pipet of the P solution <br />from 14) to 1 liter with distilled and deionized water. <br />This solution contains two micrograms of P per ml. <br />Procedure- <br />1. Add 1.OOg of 60 mesh rock or soil sample, and 20 ml of <br />Na HCO solution to a 50 ml Erlenmeyer flask. Stopper <br />the fl~sk. <br />2. Shake for 30 min., at 20°C, using a shaking speed of 2 <br />on ~ Burrell Wrist Action shaker. <br />3. Centrifuge for ].5 minutes at 2,000 R.P.M. <br />4. Pipet 10 ml of supernate into a 25 ml volumetric flask and <br />dilute to volume. Transfer to a test tube. <br />5. Slowly add 5 ml of Ammonium Molybdate solution. Mix well. <br />6. Pipet 1 ml of dilute SnC12 solution into filtering flask, <br />and mix. <br />7. Read absorbance on a spectrophotometer a 660 mu, after <br />10-20 min. <br />6-97 <br />
