Wyoming Fuel, 8505-087-135 <br />2.3 Test Or4anisms <br />Ceriodaphnia sp. were obtained from ERT's in-house culture <br />(ERT lot #063088). On the day prior to test initiation, gravid <br />females were isolated in culture water at test temperature. On <br />the day of test initiation, < 24-hour old neonates were collected <br />for use in the toxicity test. <br />2.4 Test Methods <br />The test was conducted according the Colorado Water Quality <br />Control Division's Interim Policy for Aquatic Life Biomonitoring <br />(1987). Testing was conducted in 100-mL beakers containing `_0-mL <br />of test solution. Test solutions (0, 6.25, 12.5, 25, 50, and 100 <br />percent effluent) were renewed daily with freshly prepared <br />dilutions of the initial effluent sample. At test initiation, <br />five Ceriodaphnia sp. were randomly distributed to each test <br />container and four replicates were tested per treatment. <br />Ceriodaphnia sp. were not fed during the test. The test was <br />conducted at 20 C under fluorescent lighting with a photoperiod <br />of 16 hours light and 8 hours dark. <br />In addition to the effluent test, a reference toxicant <br />(sodium dodecyl sulfate) test was also conducted with ERT's in- <br />house Ceriodaphnia sp. during the month of June, 1988 to <br />determine the sensitivity range of the test organisms. <br />2.5 Data Analysis <br />The LC50's and 95 ~ confidence limits were calculated, where <br />7 <br />
