If location has a flume or weir, collect flow data from flume or weir and <br /> record <br /> b. If no flume or weir is present and sufficient flow is present collect flow <br /> data by the following stream flow measurement procedure and the <br /> Marsh McBirney flow meter procedure. If insufficient flow is present to <br /> use Marsh McBirney flow meter estimate flow. <br /> 6. Collect a grab sample upstream from YSI probe by submerging a sampling <br /> vessel to mid depth of the stream, rinse the bottle 3 times with stream water <br /> and then collect sample. <br /> 7. Transfer sample into sample bottles that are labeled with the correct sample <br /> location, date, time, preservative and if filtration is required. <br /> a. To collect samples that do not require filtration, pour sample directly <br /> from sampling vessel. <br /> • b. To collect samples that require filtration, use decontaminated peristaltic <br /> pump. <br /> i. Attach 45 micron filter to peristaltic pump tubing. <br /> ii. Allow sample to saturate and to flow through filter <br /> (approximately 200 mL). <br /> iii. Discharge and discard approximately 100 mL of H2O from filter <br /> iv. Collect filtered sample in appropriate sample bottle(s). <br /> 8. Place samples in cooler on ice for storage. <br /> 9. Decontaminate sampling equipment. <br /> 10.
