Tubes were incubated for 1 h @ <br />370 and radioactivity decanted into a radioactive waste <br />container. The tubes were then blotted and remaining <br />radioactivity counted. The amount of radioactivity remaining <br />was considered non-specific binding. The program contained <br />on the Biogamma counter automatically subtracted the mean <br />NSB from each standard curve and unkown sample before <br />calculating percent bound <br /> <br />Ten blank tubes, i.e., tubes that did not contain 1251-T4 or T4 <br />were assayed to ensure that the gamma counter was <br />functioning properly. Distilled water was added to four Ab <br />coated tubes and incubated for 1h @ 370C. The tubes were <br />decanted, blotted and counted like other tubes. <br /> <br />All standard curve, unknown and quality control standards <br />were assayed in duplicate to control for procedural errors. For <br />each pair of samples the mean value was calculated and <br />percent error from the mean determined. The mean percent <br />error :I: 95% CI for all samples was calculated using Statview <br />II (Brainpower, Inc.) on an Apple II CX computer. A frequency <br />distribution was plotted of the percent error. <br /> <br />To determine pipetting accuracy, individuals pipetting the <br />25~1 unknown/standard curve samples and 1 ml 1251-T4 <br />samples were each required respectively to pipette 10 <br />replicate 25j.1.1 samples or 10 replicate 1 ml sample of 12 SI_ <br />T 4 into uncoated tubes. The amount of radioactivity in each <br />sample was counted in the gamma counter.
