I volunteered our lab
to conduct that analysis for the genetics panel. This report gives those results; their interpretation
is only from our lab group. Over the past several weeks, we have electrophoretically analyzed
some additional samples of razorback suckers (from Colorado via Frank Pfeiffer -- Etter Pond
and Ouray offspring and from Utah via Tim Modde -- Green River offspring) at the Illinois
Natural History Survey by Todd Kassler in Dave Philipp's lab. The purpose of these analyses
was 1) to further assess the conclusion given by Dr. Buth that the Etter Pond fish were hybrids
with flannelmouth suckers and 2) to determine the level of variation in captive adults at Vernal,
Utah for assessing the potential of constructing genetically-tagged broods for use in measuring
differential survival in flooded, riverside wetlands (e.g., Old Charley Wash).
To summarize our findings:
1) We analyzed variation at 56 loci for
a) 28 YOY RBS from Etter Pond adults
b) 6 YOY RBS from Ouray adults
c) 38 YOY RBS from Green River adults
d) 4 adult FMS from Upper Colorado River
2) Of those 56 loci:
a) 9 loci had fixed differences between all RBS and the four FMS: (ACO-1
AT-1*, CBP-I*, FH-1*, PI-Al*, IDPH-A*, mMDH-2*, DH-I*, D-2*)
b) 11 loci were fixed for a single allele among the 4 FMS, with RBS being
polymorphic and sharing the FMS allele: K-A*, K-B I*, EST-2*,
EST-3*, E T-4*, IDHP-B 1 *, LDH-C*, Mil *, MDH-B*, mMDH-1 *,
TPI-A2)
(Of those, Dr. Buth reported that K-A* and MDH-AI* had fixed differences
comparing RBS and FMS.]
c) 6 loci were fixed for a single allele among the RBS, with FMS being
polymorphic and sharing the RBS allele: (ACP-1 *, K-B2*, GPI-A2*,
IDHP-B2*, PGDH-1 *, PR T-1 *)
